Several characters had a rough time before Crisis on Infinite Earths started and Brandon Routh’s Superman himself seemed to have had an unfortunate encounter with The Joker.
Technically speaking, Crisis on Infinite Earths doesn’t name the Joker as the culprit but the implication is still very clear. When Tyler Hoechlin’s Superman encounters Brandon Routh’s Superman and he reveals what happened to his universe’s versions of Perry White, Jimmy Olsen, and Lois Lane. As it turns out, they were all killed in a gas attack by someone who is heavily implied to be the Joker. Brandon Routh’s Superman describes the person as being from Gotham and was upset that he wasn’t getting enough headlines. After telling this story, he vows to help stop the Anti-Monitor’s plans.
The death of Lois Lane and other members of the Daily Planet at the hands of the Joker was something that happened in the Kingdom Come comic. In it, the Joker flooded the Daily Planet with toxic gas killing all who were inside. Brandon Routh’s Superman may not namedrop the Joker in Crisis On Infinite Earths, but given the background, the mentioning of someone from Gotham, and the version of Superman that pays homage to Kingdom Come, it’s fairly clear who the perpetrator is.

Crisis On Infinite Earths will span two quarters and consist of Arrow, The Flash, Supergirl, Legends of Tomorrow, and Batwoman. The Arrowverse crossover will launch with three episodes in December before concluding with two additional episodes in January.
In addition to the regular cast members of each Arrowverse series, Crisis on Infinite Earths will also feature appearances by Tyler Hoechlin as Superman, Brandon Routh as Clark Kent/Superman and Ray Palmer/The Atom, LaMonica Garrett as the Anti-Monitor, Jon Cryer as Lex Luthor, Tom Cavanagh as Pariah, Elizabeth Tulloch as Lois Lane, Cress Williams as Black Lightning, Kevin Conroy as Bruce Wayne, Johnathon Schaech as Jonah Hex, Erica Durance as Lois Lane, Tom Welling as Clark Kent / Superman, Ashley Scott Will as Huntress, and Burt Ward in an undisclosed role.
